According to IGN, the Xbox 360, Wii, PC and Mac versions of the game (the latter two of which aren't out yet) have online multiplayer functionality, while the PS2 and PS3 versions do not.
All versions except the PS2, though, can connect to the GH3 community site (www.guitarhero.com) and its features.

Says here the PS3 will have online mode as well, just the PS2 will be lacking.

Makes sense. IGN is no stranger to getting things wrong; until 1 or 2 days ago, they still showed the release date for the PC and Mac versions as Oct. 29th. I just use the site because it's got info on stuff for all systems.
And no, you can't *directly* go head-to-head cross-system, but you can still compete for score, etc. cross-system via the community site.

Awesome picture and I'm also a huge Guitar Hero 3 fan. When online play actually connects, it's a blast to play. Great choice for the picture too. "Shorts" (Or Dimebag Darrell) Axel is my favorite and Squeek makes for a perfect singer :D
