The story revolves around Rin Okumura, a teenager possessing abnormal strength and endurance who, along with his younger twin brother Yukio, was raised by Father Shiro Fujimoto. Soon after Yukio departs to study at the prestigious True Cross Academy, Rin learns that he is the son of Satan, the strongest of all demons. Witnessing Shiro dying to protect him, Rin ends up doing what his guardian asked him never to do: draw the demon-slaying blade Kurikara, which restrains his demonic powers, from its scabbard. From that moment on, Rin not only gains permanent demonic features like fangs, pointed ears and a tail, but also the power to ignite himself into blue flames that can destroy anything they touch.

At Shiro's funeral, Rin has an encounter with Mephisto Pheles, who agrees with Rin's wish to become an Exorcist like his guardian to take revenge on Satan. Rin enrolls at the True Cross Academy, which is actually the Japanese branch of an international organization dedicated to protect the human realm, Assiah, from demons and other creatures from the demon realm, Gehenna. Rin takes part in a special Exorcist course, the existence of which is known only by a few. Much to his surprise, he finds that his brother Yukio is already a veteran Exorcist and will be one of his teachers.

Shura possesses long, red hair with yellow ends that resemble flames. Her eyes are green in the manga, yet purple in the anime, and she has a curvaceous figure (she has an F-cup). She is usually seen with a (very revealing) bikini top over her endowed breasts, along with shorts and sometimes with stockings as the bottom part of her outfit or also wearing a denim jacket. After she is assigned to become a teacher at the Exorcist Cram School, Shura starts wearing an unbuttoned school jacket and a tie. She also carries a license that states her name and position, as well as a key with coat of arms on it, which symbolizes her position. She also has markings on her body where she, after saying "Devour the Seven Princesses, slay the Serpent", can get her sword from. The area where she attains the sword from is on her upper-chest. Shura has a rather laid-back personality and uses slang; using yer instead of your. She is also noted to be rather blunt, speaking freely about sensitive topics, as well as not being hesitant in pointing out others' flaws. Even with her superiors, she is not afraid to challenge them, which on a few occasions resulted in physical conflict.

Like Rin, she often slacks off, and would rather drink and let others do work. However, when needed, Shura can be serious and focused, as seen when she duels Amaimon and being able to depict the fact that Rin's flames take over him because of his fear. She is also fiercely loyal to Father Fujimoto, despite denying his ways. Insulting or playing around with Shura can lead to being punished. Shura is currently hiding her true age by stating to everyone that she is 18, when Yukio has alluded to the fact that she is 26. Little is known about Shura's past, except that Father Fujimoto saved her as a child from the path of evil. He also took her as his apprentice, and passed on his knowledge and skill to her.

She eventually earned the title Meister as both a Knight and a Doctor. Early in Rin's life, Shiro requested in their last meeting that if he were to die, she teach Rin to wield Koumaken, saying that no one knew more about demon swords than she did. She stubbornly refused, lashing out that he was throwing away his life for the son of Satan.
