Ya know, most people dont even try to do something like this, an experimental action pose that's made even more difficult by the character's unusual anatomy, its kinda hard to conceptualize and draw in a believable way.
Personally, I'd angle her front foot forward, as she'll be needing her soles pointed more at the ground so that she can land on it easily when she takes that step; I'd straighten out her front leg a bit, as she needs that leg to be able to support her considerable weight to be able to take that step without collapsing; I'd keep her step height fairly low to the ground, since her tremendous weight would make large sprint-like bounds exhausting, even with a lifetime of weight training; I'd make her body a little less twisted, as she's trying to run forward and here it looks like she's going off to her right a bit, since the body tends to stay fairly oriented forward while running; also, finally, sort of, I'd keep her belly more-or-less in front, since even though its soft and bouncy, its also heavy, which makes it difficult to move violently, and held with a lot of connective tissue, so to keep it from just going wherever it wants.
A thought for what kind of manner of running someone this bulky and heavy might use, I'd think more of an energetic shuffle-jog sort of thing would be more her speed, something that wouldnt need her to spend so much energy lifting her body so far off the ground, and isnt so high impact that it becomes to painful to run, what with her skin repeatedly trying to keep the better side of a ton's worth of fat from spilling through, yet still moves her body forward faster than just walking. Though, this also implies she's put a lot of thought into the most efficient way to run, which I doubt, since she looks like the type not to consider running as something other than torture, which then it would make sense for her to try to break into a sprint, do something awkward and tiring and painful, then-- I think I'm getting a little off topic here.
Regardless of how accurate you drew the pose, you went out there, did something non-trivial, and have something nice to show for it. That's what makes a good artist, the willingness to do something they havent done (well enough) before, so good on you for that.

Yeah, coming up with a good pose that not only showed motion, but also her sheer girth, was pretty tricky; especially since like you said, most people don't really draw characters of Puff's build in situations like this. ^_^;
Despite that however, I think it came out pretty good overall; though the next time I try to do something like this again, I'll keep your advice in consideration. =3
