And to be honest with Muggshot, Sly and Murray had built muscles, but only by chance.
First, Muggshot assaulting Bentley and stealing Dawkins' journal is pretty much out of payback for the turtle insulting the bulldog's mother back in Holland.
Secondly, Sly and Murray were unable to hurt Muggshot with their normal attacks in Mesa City and Holland, respectively, but now thanks to the turquoise stone's magic making Sly and Murray bigger, tougher, and stronger than ever, they're now able to actually hurt Muggshot in a fistfight since they're even huger and tankier than Muggshot and Jean Bison.
