At least someone made Jenka's Nightmare, a sequel fangame that even give you some extras if you transfer your save datas from CS. You should check it out if you like the first game !

Cave Story was one amazing exercice in worldbuilding, and after you beat it you're left with the feeling that there's so much more to explore and uncover about the island... Actually forget a new game, I would be just as satisfied with a comics or a movie !
The Switch version of the game was recently updated with a 2P mode, I hope it means the franchise isn't forgotten yet !
